TGF-β superfamily

Transforming growth factor beta (TGF-β) superfamily encompasses a range of proteins including TGF-β, growth differentiation factors (GDFs) and bone morphogenetic proteins (BMPs), glial-derived neurotrophic factor (GDNF), inhibins, activins, nodal, lefty, and Mülllerian inhibiting substance (MIS).

Shared among these members are dimeric structures and receptor complexes composed of both type I and type II receptor subunits, each containing serine/threonine kinase domains. Upon ligand binding, the type II receptor phosphorylates and activates the type I receptor, initiating a signaling cascade reliant on Smad proteins that ultimately regulates gene expression.

This superfamily of proteins plays diverse roles in embryonic development, morphogenesis, adult stem cell differentiation, wound healing, immune regulation, cancer and inflammation.
