Recombinant GDF-11 activity was determined using a Smad reporter assay in transfected HEK293 cells. HEK293 cells were treated in triplicate with a serial dilution of GDF-11 for 6 hours. Firefly activity was measured and normalized to the control Renilla luciferase activity. Data from Qk126 lot 204827. EC50 = 0.69 ng/ml (27.6 pM)

Recombinant GDF-11 (BMP-11) at approximately 12.5 kDa (monomer) in reduced (R) and approximately 25 kDa non-reduced (NR) conditions. No contaminating protein bands are present. The purified recombinant protein (1.5 µg) was resolved using 15% w/v SDS-PAGE in reduced (+β-mercaptoethanol, R) and non-reduced (NR) conditions and stained with Coomassie Brilliant Blue R250. Data from Qk126 lot #204827.

Product background

Bone morphogenetic proteins (BMPs) and growth differentiation factors (GDFs) belong to the TGF-β superfamily of structurally related signaling proteins. BMPs and GDFs form a family of cystine-knot cytokines which display morphogenetic activities in the development of a wide range of tissues. Existing as homodimers or heterodimers, they have tissue specific functions including the development of bone, kidney, and cardiac tissues [1].

GDF-11/BMP-11 has a high degree of homology to GDF-8/myostatin [1, 2], and is a key regulator of cell growth and differentiation during muscular and neural development. Both GDF-11/BMP-11 and GDF-8/myostatin bind to the activin type 2 receptors and recruit ALK4 or ALK5 to activate Smad2/3 signaling [2]. GDF-11/BMP-11 has also been shown to recruit ALK7 or ALK1 to activate Smad1/5/9 which may be responsible for the difference in signaling and developmental responses [2].

Circulating GDF-11/BMP-11 levels decrease with age, highlighting it as a possible therapeutic in age-related diseases [3]. Recombinant GDF-11/BMP-11 has pro-myogenic effects in skeletal muscle and anti-hypertrophic effects in the heart and beneficial effects on neurogenesis and neuronal function [4] leading to therapeutic possibilities in the treatment of stroke [3]. It increases proliferation in aged satellite cells, an effect not seen with GDF-8/myostatin or TGF-β1 [4]. In studies using mesenchymal stem cells (MSCs) recombinant GDF-11/BMP-11 dose-dependently decreased adipogenesis by activating Smad2/3‐dependent TGF‐beta signalling pathway, a process which could be reversed with ALK4/5 inhibitors [5]. GDF-11/BMP-11 has also been shown to decrease both osteoblast [6] and chondrocyte differentiation [7], making it a potential therapeutic target for osteoporosis and fracture healing.

FAQ

What is GDF-11/BMP-11?

GDF-11 / BMP-11 is a TGF-β superfamily protein with an essential role in body patterning during development.

Where is GDF-11/BMP-1 found?

During development GDF-11 is highly expressed in the tail bud; it is also expressed widely in adult tissues including the nervous system, muscle, kidneys and pancreas.

Is GDF-11/BMP-11 a cytokine?

Yes, as a member of the TGF-β superfamily, GDF-11 / BMP-11 is classed as a cytokine.

What does the GDF-11/BMP-11 gene do?

The gene GDF11 codes for growth differentiation factor 11 (GDF-11) a protein also known as bone morphogenetic protein 11 (BMP-11).

What does GDF-11/BMP-11 bind to?

GDF-11/BMP-11 binds to activin type II receptors and induces phosphorylation of Smad2.

What is the function of the GDF-11/BMP-11 receptor?

Activin receptors activate the Smad signaling pathways. Smad proteins can also be associated with various tissue-specific transcription factors, which mediate a range of processes, including pluripotency.

How is GDF-11/BMP-11 used in cell culture?

GDF-11/BMP-11 has been used in the study of adipogenesis, neurogenesis, and osteogenesis.

Does GDF-11/BMP-11 have therapeutic use?

GDF-11/BMP-11 is of therapeutic interest in age-related diseases as it influences muscle, neural and bone healing and degeneration.

Special guidance for reconstitution of TGF beta family including activins, BMPs and GDFs

TGF beta family proteins and other growth factors can be very poorly soluble in physiological solutions.  Please follow the handling guidance for lyophilized cytokines below to minimize loss of protein due to precipitation or adsorption to plastic.  We advise storing the recombinant protein at very low pH before dilution in cell culture media or final working solutions. Low pH will also assist in maintaining the correct disulfide structure of the protein by minimizing disulfide bond exchange reactions.

  • Resuspension in physiological buffers may cause precipitation of stock solutions, hence we recommend dissolving our lyophilized cytokines in 10 mM HCl (1:1000 dilution of concentrated HCl) while keeping the protein concentration at 50 µg/ml or above, in order to avoid loss by adsorption to plasticware.
  • To ensure you recover all of the protein, let the sample sit for a few minutes with the solubilization buffer at room temperature and pipette gently up and down (avoid foaming).
  • Rinse the tube with some more 10 mM HCl and pool with the rest.
  • The protein is tolerant of some freeze and thaw cycles, but as always with proteins, it is better to aliquot and store frozen.
  • Our proteins are supplied carrier-protein free.  If compatible with your work, add carrier protein of your choice such as BSA, HSA or gelatin to further minimize loss by adsorption.
  • Store in -80°C for long term storage. -20°C for short-term.

Every effort is made to ensure samples are sterile; however, we recommend sterile filtering after dilution in media or the final working solution.
