Interleukins

IL superfamily is a large group of cytokines, including proteins known as interleukins and interleukin receptors. Members of this superfamily play pivotal roles in diverse biological processes such as immunity, inflammation, and hematopoiesis. Subgroups within the interleukin superfamily include the IL-1, IL-6, IL-10, IL-12, and IL-17 subfamilies, along with additional interleukins and receptors.

Interleukins are a large group of cytokines with immunomodulatory functions that elicit a wide variety of responses in cells and tissues.  Interleukin proteins play an important role in growth, differentiation and activation of cells in the hematopoietic and  immune systems.

Interleukins also play a crucial role in orchestrating the body’s physiological response to infection, while also playing a significant part in the functional alterations associated with specific diseases or disorders. Given their pivotal role, it has become clear that interleukins represent a group of proteins with considerable potential as therapeutic targets.

Interleukin superfamily can be sub-divided into several major groups, such as:

– Interleukin-1 (IL-1) family

Interleukin-1 (IL-1) family cytokines are a subgroup of the interleukin superfamily of cytokines. It is group of 11 proinflammatory cytokines which regulate inflammation. This group consists of IL-1α, IL-1β, IL-36α, IL-36β, IL-36γ, IL-36RA, IL-37, IL-38, and IL-1RA which share a common genetic lineage, plus IL-18 and IL-33 which are included due to structural similarly.

– Class I helical cytokines (IL-4-like and IL-6/12-like)

A large family of signaling molecules characterized by a three-dimensional fold consisting of a bundle of four alpha-helices. This family includes IL-2, IL-15, IL-4, IL-7, IL-9, and IL-21 and EPO.

– Class II helical cytokines (IL-10-like and IL-28-like)

A family of cytokines characterized by their alpha-helical structure and their ability to bind to and signal through class II cytokine receptors. This family includes various interferons (IFNs) and the interleukin-10 (IL-10) family of cytokines.

– Interleukin-6 (IL-6) family

IL-6 family play roles in various processes, including inflammation, immune cell differentiation, and hematopoiesis. This family includes IL-6, IL-11, CNTF, LIF, OSM, CT-1, CLC, and IL-27.
