Interleukin 21 (IL-21) is a cytokine produced by natural killer (NK) cells and some T cells. IL-21 has broad immunological functions and can regulate NK cells, macrophages, and dendritic cells, as well as playing a role in promoting B cell and antibody responses. Recombinant IL-21 activity was determined using a STAT3 luciferase reporter assay in HEK293 cells co-transfected with the IL-21 receptors (IL-21R and IL-2Rγ). Transfected HEK293 cells were treated in triplicate with a serial dilution of IL-21 for 18 hours. Firefly activity was measured and normalized to the control Renilla luciferase activity. Data from Qk129 lot #204831. EC50 = 4.17 ng/ml (0.27 nM).
Recombinant IL-21 migrates at approximately 15.3 kDa (monomer) in reduced (R) and non-reduced (NR) conditions. No contaminating protein bands are present. The purified recombinant protein (3 µg) was resolved using 15% w/v SDS-PAGE in reduced (+β-mercaptoethanol, R) and non-reduced (NR) conditions and stained with Coomassie Brilliant Blue R250. Interleukin-21 (IL-21) is a potent immunoregulatory cytokine produced by activated Natural Killer Cells (NKT), T helper (Th) 17 and T follicular helper (TFH) cells [1]. IL-21 expression is induced by T cell receptor signaling, and by other cytokines such as IL-1β, IL-6 and IL-27 [1]. IL-21 protein exerts its biological effects through a heterodimeric receptor composed of the IL-21-specific IL-21 receptor (IL-21R) and γ-chain (γC), which is also a receptor for IL-2, IL-4, IL-7, IL-9, and IL-15 [2]. Upon binding to its receptor IL-21 signals through the Jak-STAT signaling pathway as well as the MAPK, and PI3K pathways [1, 2].
IL-21 stimulates the activation, proliferation, and survival of CD8+ T cells and NKT cells and promotes Th17 cell polarization. In combination with TGF-β1, IL-21 promotes the differentiation of human naive CD4+ T cells into Th17 cells [3]. In addition to its role in T cell biology, IL-21 also plays a critical role in B cell activation, proliferation, differentiation, apoptosis, antibody production and differentiation into plasma cells [1, 2, 4]. It is also required for the migration of dendritic cells to draining lymph nodes.
