Product background
LY294002 is a broad-spectrum inhibitor of phosphoinositide 3-kinases (PI3Ks), inhibiting PI3Kα, PI3Kδ, and PI3Kβ (IC50=0.5/0.57/0.97 μM). LY294002 is also an inhibitor of DNA-PK (IC50=1.4 μM) and CK2 (IC50=98 nM) but not PI4K or MAP kinases [1]. PI3K is activated through growth factor stimulation leading to an increase into the secondary messenger phosphatidylinositol (3,4,5)-trisphosphate (PIP3) which recruits pleckstrin homology (PH) domain containing proteins. Through PIP3, PI3Ks activate major cellular signaling pathways such as Akt, GSK3α/β, mTORC1 and PDK1 [2, 3] and cross talk with the WNT/β-catenin, NF-κB and TGF-β signaling pathways [3]. Through activation of multiple downstream signaling pathways PIP3 stimulates cellular proliferation, survival, motility and metabolism and other processes considered oncogenic [2, 3].
PI3K signaling is important in the regulation of cancer stemness and the maintenance of undifferentiated embryonic stem cells (ESCs) and reprogramming of induced pluripotent stem cells (iPSCs) [3, 4]. In mesenchymal stem cells (MSCs), PI3K plays a role in osteogenic and adipogenic differentiation in combination with PDGF and TGF-β signaling pathways [4]. Inhibition of PI3K with LY294002 inhibits MSC proliferation and promotes neural differentiation, it can also be used to promote differentiation of human endometrial stem cells (hEnSCs) into motor neuron-like cells [5]. LY294002 can be used in ESC to inhibit the IGF/CK2 pathway to promote cardiomyocyte differentiation [6].
