SB431542 is a potent and selective inhibitor of ALK4, ALK5 and ALK7 receptors which activate the TGF-β/activin/nodal pathway. This pathway is essential for maintaining pluripotency in embryonic (ESC) and induced pluripotent stem cells (iPSC). In cell culture, SB431542 can be used for induced differentiation of ESC and iPSC into mesenchymal lineages, cardiac and neural differentiation.

Product background

SB431542 is a potent and selective inhibitor of TGF-β type I Receptors ALK5 (IC50 = 94 nM) and ALK4 (IC50 = 140 nM) and ALK7 [1,2]. SB431542 does not have an effect on BMP, ERK, JNK, or p38 MAP kinase pathways or signaling pathways activated in response to serum [2]. TGF−β superfamily ligands including activins and nodal signal through these receptors, mediating essential cellular processes related to development, would healing and immune regulation [2, 3].

In embryonic stem cells (ESCs) activin/nodal signaling in combination with FGF-2 maintains pluripotency through the ALK4/5/7 receptor pathway, addition of SB431542 reduces expression of pluripotency marker Oct-4 [4]. SB431542 can also be used to induce differentiation of both ESCs and induced pluripotent stem cells (iPSCs) into mesenchymal stem cells (MSCs) [5]. As part of cell culture media, SB431542 enhances differentiation into neurons [6, 7], astrocytes and cardiomyocytes [7].
