Interleukin-34 (IL-34) is a cytokine involved in regulating the differentiation, survival, and activation of monocytes, macrophages, and osteoclasts. IL-34 plays a key role for the development and maintenance of tissue-resident macrophages such as Langerhans cells in the skin and microglia in the brain.
Qkine IL-34 PLUS™ is animal origin-free and has comparable bioactivity to the most bioactive mammalian expressed alternative supplier protein.
Qkine IL-34 PLUS™ bioactivity
- Qkine IL-34 PLUS™ (Qk091) was bioactive in a quantitative luciferase assay with EC50 of 7.41 ng/ml (0.29 nM).
- This was as bioactive as mammalian expressed IL-34 from supplier A, which had an EC50 of 12.6 ng/ml (0.48 nM).
- Qkine IL-34 was more bioactive than mammalian expressed IL-34 from supplier B which had an EC50 of 179 ng/ml (3.4 nM).
- A bacterially expressed IL-34 was also tested and found to not be bioactive.
Qkine IL-34 PLUS was as bioactive as a mammalian cell-expressed IL-34 from an alternative supplier and more bioactive than others. IL-34 activity was determined using an SRE reporter assay in HEK293 cells co-transfected with the CSF1R receptor. Transfected HEK293 cells were treated in triplicate with a serial dilution of Qkine IL-34 PLUS (Qk091, green), R&D Systems (5265-IL) mammalian cell-expressed IL-34 (Supplier A, black), Peprotech (200-34) mammalian cell-expressed IL-34 (Supplier B, blue) or Creative Biomart (IL34-31H) bacterially-expressed IL-34 (Supplier E, pink) for 3 hours. Firefly activity was measured and normalized to the control Renilla luciferase activity.
