Qkine platelet derived growth factor BB (PDGF-BB, Qk044) is more biologically active than a comparable alternative supplier protein
PDGF-BB is a member of the PDGF family, it is a homodimer of PDGF-B peptide chains. Its is a potent mitogen expressed by platelets which stimulates proliferation and angiogenesis and is involved in tissue repair. PDGF-BB increases stem cell marker expression and can be used in osteogenic and neural stem cell differentiation.
Qkine PDGF-BB (Qk044) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.
Qkine PDGF-BB bioactivity
• PDGF-BB (Qk044) was bioactive in a quantitative luciferase assay with an EC50 of 12.5 ng/ml (0.5 nM).
• This was more bioactive than an alternative supplier, which had an EC50 of 25.85 ng/ml (1.03 nM).
Qkine PDGF-BB was found to be more bioactive than PDGF-BB from an alternative major supplier. Bioactivity was determined using the NFAT luciferase reporter assay in HEK293T cells co-transfected with PDGFBR. Cells were treated in triplicate with a serial dilution of Qkine PDGF-BB (Qk044, green) or Peprotech PDGF-BB (100-14B-10UG, Supplier B, black) for 6 hours. Protein concentrations were measured before use (Peprotech vial recovery 83%, Qkine 116%). Firefly luciferase activity was measured and normalized to control Renilla luciferase activity. This bioactivity comparison demonstrates that Qkine PDGF-BB was more bioactive than PDGF-BB from an alternative major supplier. Qkine recombinant proteins are more cost effective and have the advantage of having high and consistent vial recovery. They are highly pure and animal origin-free, giving lot-to-lot consistency in bioactivity for long-term reproducible culture of stem cells and organoids.
