Technote | IL-1 beta (Qk101)
Qkine interleukin-1 beta (IL-1β) is slightly more biologically active than a comparable alternative supplier protein
Recombinant human interleukin-1 beta (IL-1β) is a pro-inflammatory cytokine and a potent mediator of inflammation and immune responses. In cell culture, IL-1β is used to stimulate and activate immune cells (macrophages and T cells), maintain hematopoietic progenitor cells, and modulate the differentiation of mesenchymal stem cells.
Qkine IL-1 beta (Qk101) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.
Qkine IL-1 beta bioactivity
- Qkine IL-1 beta (Qk101) was bioactive in a quantitative luciferase assay with EC50 of 129 ng/ml.
- This was slightly more bioactive than IL-1 beta from an alternative supplier, which had an EC50 of 232 ng/ml.
Quantitative luciferase reporter assay shows Qkine IL-1 beta bioactivity. Bioactivity was determined using a NFkB quantitative luciferase assay. HEK293T luciferase reporter cells were treated in triplicate with a serial dilution of Qkine IL-1 beta (Qk101, green) or Peprotech (120-01B, supplier B, black) for 3 hours. Protein concentrations were measured before use (Peprotech vial recovery 90%, Qkine 112%). Firefly luciferase activity is measured and normalized to control Renilla luciferase activity. Data from Qk101 lot #204581.
Qkine IL-1 beta has the advantage of being more cost effective, highly pure and animal origin-free, giving batch-to-batch consistency in bioactivity for long-term reproducible culture of hematopoietic progenitor and other stem cells.
