Qkine granulocyte macrophage colony-stimulating factor (GM-CSF) is stable and bioactive up to 28 days in cell culture conditions

Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a hematopoietic growth factor involved in differentiating and activating monocytes and granulocytes. It is also involved in developing erythroid cells, megakaryocytes, and keratinocytes. GM-CSF is commonly used in cell culture to stimulate the differentiation and maturation of human induced pluripotent stem cells or peripheral blood cells to myeloid lineages.

Qkine GM-CSF (Qk076) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.

• After 14 days Qkine GM-CSF (Qk076) stimulated proliferation of TF-1 human myeloid leukemia cells with the same potency as fresh GM-CSF, with an EC50 at day 0 of 68 pg/ml and 80 pg/ml at day 14.
• GM-CSF was still bioactive after 28 days, with an EC50 at day 0 of 71 pg/ml and 73 pg/ml at day 28.
• No degradation of protein can be seen in an SDS-PAGE gel after 14 or 28 days.

Bioactivity of Qkine GM-CSF at 14 days. Bioactivity was determined using  proliferation of TF-1 human myeloid leukaemia cells. Cells were treated in triplicate for 48 hours with a serial dilution of GM-CSF which had been pre-incubated in conditioned media at 37°C for 14 days. Cell viability was measured using CellTiter-Glo (Promega) luminescence assay.

Bioactivity of Qkine GM-CSF at 28 days. Bioactivity was determined using  proliferation of TF-1 human myeloid leukaemia cells. Stability of Qkine GM-CSF protein. SDS-PAGE of recombinant GM-CSF showed that protein was not degraded when incubated in HEK293T conditioned media for 14 or 28 days at 37°C.

Qkine GM-CSF (Qk076) is stable and bioactive under cell culture conditions for up to 28 days. Qkine GM-CSF is highly pure and animal origin-free, giving batch-to-batch consistency in bioactivity for long-term reproducible culture of myeloid cells and other relevant hematopoietic cells.
