Qkine interleukin-6 (IL-6) is stable and bioactive up to 28 days in cell culture conditions
Interleukin 6 is a multifunctional cytokine that regulates immune responses and inflammation. It is produced by various cells, including immune cells such as T cells and macrophages, as well as non-immune cells like fibroblasts and endothelial cells. IL-6 is used for the culture of hematopoietic stem cells (HSC) and other relevant cells.
Qkine IL-6 (Qk093) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.
Qkine IL-6 stability
- After 14 days Qkine IL-6 (Qk093) bioactivity was equivalent to fresh IL-6, with an EC50 at day 0 of 3.2 ng/ml and 2.5 ng/ml at day 14.
- IL-6 was still bioactive after 28 days, with an EC50 at day 0 of 3.6 ng/ml and 3.5 ng/ml at day 28.
- No degradation of protein can be seen in an SDS-PAGE gel after 14 or 28 days.
Bioactivity of Qkine IL-6 at 14 days. Bioactivity was determined using the IL-6-responsive firefly luciferase reporter assay. Transfected HEK293T cells were treated in triplicate for 24 hours with a serial dilution of IL-6 which had been pre-incubated in conditioned media for 14 days. Firefly luciferase activity was measured and normalised to the control Renilla luciferase activity.
Bioactivity of Qkine IL-6 at 28 days. Bioactivity was determined using the IL-6-responsive firefly luciferase reporter assay. Stability of Qkine IL-6 protein. SDS-PAGE of recombinant IL-6 showed the protein was not degraded when incubated in HEK293T conditioned media for 14 or 28 days at 37°C.
Qkine IL-6 (Qk093) is stable and bioactive under cell culture conditions for up to 28 days. Qkine IL-6 is highly pure and animal origin-free, giving batch-to-batch consistency in bioactivity for long-term reproducible culture of myeloid cells and other relevant hematopoietic cells.
