Technote | GDF-5 (Qk070)
Qkine human growth differentiation factor 5 (GDF-5) bioactivity is comparable to an alternative supplier protein
Recombinant human GDF-5 protein is commonly used for the differentiation and maintenance of induced pluripotent stem cells, embryonic stem cells, or bone marrow-derived mesenchymal stem cells into osteoblasts and chondrocytes.
Qkine GDF-5 (Qk070) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.
Qkine GDF-5 Bioactivity
- GDF-5 (Qk070) was found to be bioactive in a quantitative luciferase reporter assay, with an EC50 of 1.1 ng/ml.
- This was comparable to GDF-5 from an alternative supplier, which had an EC50 of 1.0 ng/ml.
Qkine GDF-5 was as bioactive as GDF-5 from an alternative major supplier. Bioactivity was determined using a GDF-5-responsive firefly luciferase reporter assay. Stably transfected HEK293T cells were treated in triplicate with a serial dilution of Qkine GDF-5 (Qk070, green) or Peprotech (120-01, Supplier B, black) for 24 hours. Protein concentrations were measured before use (Peprotech vial recovery 108%, Qkine 107%). Firefly luciferase activity was measured and normalised to the control Renilla luciferase activity. Data from Qk070 lot #204559.
Qkine recombinant proteins are highly pure and animal origin-free, giving lot-to-lot consistency in bioactivity for long-term reproducible culture of stem cells and organoids.
